A muscular path to quiescence

چکیده

A muscular path to quiescence uscle stem cells called satellite cells can pull back from the brink, according to Zammit et al. (page 347). When activated by muscle damage, the cells divide and start uniformly down a differentiation path. Only at the last minute do some of the cells split off to revert to a quiescent, stem cell phenotype. Satellite cells sit quietly underneath the basal lamina of myofibers until a muscle is damaged. Either mitogens leaking into a damaged muscle or signals from a deinnervated muscle may wake up the satellite cells, which then divide and supply new cells that fuse to repair or replace damaged myofibers. Zammit et al. isolated intact myofibers and placed them in mitogen-laden cultures to activate the satellite cells. The activated cells, which when quiescent make the transcription factor Pax7 but not the muscle determinant MyoD, turned on MyoD. Most of the doubly positive cells then turned off Pax7 and differentiated, but some turned off MyoD instead and reverted to a quiescent phenotype. The Pax7 ϩ , MyoD Ϫ cells could not have arisen from a persistent, fast-dividing pool of Pax7 ϩ , MyoD Ϫ cells because at intermediate times such cells were found on only a few myofibers. The authors also noted that recently replicated cell pairs may have provided evidence for a recent switch, as in some cases only one of the M Do yeast apoptose? uicide in a unicellular organism might sound like a lousy idea, but there are several possible justifications. Budding yeast cells that are sickly, mating-incompetent, or stuck in the middle of a colony might do better to go out with a messy bang, thus releasing all their nutrients for their nearby relatives, than to fade away with energy sources locked behind a cell wall. Yeast do die of causes other than old age, but it remains unclear whether this death is like mammalian apoptosis, and to what extent it is controlled rather than a direct result of an insult to the cell. Apoptosis was put forward as a possibility when a caspase-like protein, Yca1p, was discovered , and dying cells were found to bind a caspase substrate. S But on page 311, Wysocki and Kron bring these conclusions, and the whole concept of yeast apoptosis, into doubt. Using yeast that are irreversibly arrested because of exposed telomeres, they find that it is the fluorophore, not the attached …

Distinct Mechanisms Underlie Quiescence during Two Caenorhabditis elegans Sleep-Like States.

Electrophysiological recordings have enabled identification of physiologically distinct yet behaviorally similar states of mammalian sleep. In contrast, sleep in nonmammals has generally been identified behaviorally and therefore regarded as a physiologically uniform state characterized by quiescence of feeding and locomotion, reduced responsiveness, and rapid reversibility. The nematode Caenor...

Notch signaling deficiency underlies age-dependent depletion of satellite cells in muscular dystrophy

Duchenne muscular dystrophy (DMD) is a devastating disease characterized by muscle wasting, loss of mobility and death in early adulthood. Satellite cells are muscle-resident stem cells responsible for the repair and regeneration of damaged muscles. One pathological feature of DMD is the progressive depletion of satellite cells, leading to the failure of muscle repair. Here, we attempted to exp...

Nitric oxide donors improve prednisone effects on muscular dystrophy in the mdx mouse diaphragm.

In Duchenne muscular dystrophy (DMD), palliative glucocorticoid therapy can produce myopathy or calcification. Since increased nitric oxide synthase activity in dystrophic mice promotes regeneration, the outcome of two nitric oxide (NO) donor drugs, MyoNovin (M) and isosorbide dinitrate (I), on the effectiveness of the anti-inflammatory drug prednisone (P) in alleviating progression of dystroph...

Ecophysiological responses of the seminal vesicle of Libyan jird (Meriones libycus) to the Saharan conditions: histological, morphometric and immunohistochemical analysis.

The Libyan jird (Meriones libycus) is a nocturnal Saharan Rodent submitted to a seasonal cycle of reproduction characterized by a short active period during spring and beginning of summer, and a long phase of sexual quiescence from the end of summer until the end of winter. During this cycle, the male reproductive organs, and more particularly seminal vesicles, experience some important weight ...

Possible dual roles for prostacyclin in human pregnancy and labor.

During pregnancy, the muscular layer of the uterine wall known as the myometrium, which is composed mainly of smooth muscle cells, is maintained in a state of relative quiescence. A switch from myometrial quiescence to myometrial activation is required to establish uterine contractions during labor. Researchers have long been perplexed by the fact that the major prostaglandin produced by the ut...
